The elements or compounds that are used to prepare the biuret reagent include sodium hydroxide, hydrated copper with potassium sodium tartrate. The reason for adding the 3rd compound mentioned is the stability of the cupric ions in the reagent. Once these compounds are added in a fixed proportion, the biuret reagent is prepared for the test. Make sure that you take important precautions while preparing the reagent and also while performing the rest of the test. Once you have all the things including the reagent, we can proceed to perform the biuret method.

The procedure for determination of plasma proteins by biuret method is a very simple one. It takes only few steps to do the procedure and then we can get the desired results.   1. In the first step, a base is taken which could be either potassium or potassium hydroxide and its amount must be 1%.
  2. Next step is to treat this base with a water sample. Also, a few drops of copper sulfate must be added during the procedure.
  3. In the step three, you start getting results just after performing the above two steps.

After performing the first two steps in the test, if the color of the solution turns to purple, it means that the proteins are present in the solution. If the purple color doesn’t appear, we are sorry to say that your test is negative. The purple color is the major indicator for the presence of proteins and this gives an easy indication really.

  • Make sure that you have measured amount of all the chemicals correctly.
  • While making the reagent, taking notice of the proportions is must.
